作用機(jī)制
本品為*核苷類似物,對(duì)乙肝病毒(HBV)多聚酶具有抑制作用。它能夠通過(guò)磷酸化成為具有活性的三磷酸鹽,三磷酸鹽在細(xì)胞內(nèi)的半衰期為15小時(shí)。通過(guò)與HBV多聚酶的天然底物三磷酸脫氧*核苷競(jìng)爭(zhēng),
*三磷酸鹽能抑制病毒多聚酶(逆轉(zhuǎn)錄酶)的所有三種活性:
(1)HBV多聚酶的啟動(dòng);
(2)前基因組mRNA逆轉(zhuǎn)錄負(fù)鏈的形成;
(3)HBV DNA正鏈的合成。*三磷酸鹽對(duì)HBV DNA多聚酶的抑制常數(shù)(Ki)為0.0012μM。*三磷酸鹽對(duì)細(xì)胞的α、β、δDNA多聚酶和線粒體γDNA多聚酶抑制作用較弱,Ki值為18至于160μM。
